While glucosamine is beneficial, it also has disadvantages. Those who are allergic to shellfish should not take glucosamine because it is manufactured from naturally occurring shellfish and some of the allergens can be present in the supplement. Also, another drawback of glucosamine is that it may increase the severity of diabetes in those who have it. As with any other medication and supplement, glucosamine may cause harsh side effects. Side effects may include nausea, diarrhea, heartburn, drowsiness, skin rash and headache.

There has not been enough research to recommend glucosamine for use with children under the age of 18. MSM, or methylsulfonylmethane, is sometimes packaged with glucosamine and some research, according to MayoClinic.com, has shown a possible relationship between it and autism.
